There could be several reasons causing a conflict between the houses of Montague and Capulet. The first one could be a clash of interests caused by their beliefs. Religion has always been a source of trouble. To believe in one god or another, or having religious habits that are different from others, still separates cultures today. Since the piece played in the late ages of the sixteenth century (Renaissance), the majority fo the people living in Verona were Catholics. It might be possible that either one, Montagues or Capulets, had changed their beliefs due to the Protestant Reformation, which also took place in the 16th century.
The other reason could simply be money. We know that today's family conflicts are mainly about money. Why wouldn't it be the same back then? One family has more money than the other does, and voilà: we have our fight that keeps on dividing the two families over generations. Back then it might even had been the difference of social classes, that made the two houses fight each other.
